Abstract
The aim of this study was to produce chimeric bacterial protein conjugates, to separate them by sodium dodecyl sulphate- polyacrylamide gel electrophoresis (SDS-PAGE) and study their reactivities with human serum immunoglobulins by immunoblot analysis. Universal conjugates: protein LA-peroxidase (SpLA-HRP), protein LG-peroxidase (SpLG-HRP), protein AG-peroxidase (SpAG-HRP), protein LAG-peroxidase (SpLAG-HRP) and protein LAG-anti-IgY-HRP were prepared by the periodate method. In this study, antibodies against the fragment 254-274 from gp120 of human immunodeficiency virus (HIV) were produced in cats, rats and chicken and human samples were used as positive and negative controls in the ELISA. These anti- HIV antibodies developed in animals could be used as reagents in the preparation of diagnostic kits. The ELISA for anti-gp120 antibodies demonstrates that SpLAG-anti-IgY-HRP conjugate is the most universal among available conjugate reagents, which can be used in the immunodiagnosis in both avian and mammalian species.
Biography
Angel Justiz Vaillant has completed his M.D and Ph.D. at the age of 35 years from The Higher Institute of Medical Sciences, Santiago de Cuba and University of the West Indies, Mona campus (UWI) respectively. He is Lecturer in Immunology at UWI, St Augustine. He has published more than 15 papers in reputed journals and has been serving as a technical editor for various journals.
