Quantitative real-time PCR compared with pp65 antigen detection for Human Cytomegalovirus (HCMV) in isfahan province

Abstract
Human cytomegalovirus: HCMV is a member of herpes virus family and its early diagnosis could be very critical in kidney transplant patients. It is because of the wide range of HCMV infection clinical appearances from tissue rejection to extremely death. Th e goal of this study is the comparison of two common methods of HCMV monitoring (real-time PCR and pp65 antigen assay) to discover the accuracy of these methods. In this study, we extract the genomic DNA of 135 kidney transplant patients from their total blood in the Isfahan privince. HCMV monitoring was done according to Taq Man probe scanning and pp65 surface antigen detection using real-time PCR and pp65 antigen assay respectively. Th e results of our study indicated that real-time PCR shows positive results in 36 patients (27%) but in pp65 antigen assay 29 patients (21%) detected as HCMV positive. It was also clear the maximum infection of HCMV belong to spring (28%) and the minimum rate of virus infection appear in winter (16%) but no signifi cant association detected between HCMV infection and seasonal patterns. Our results indicated that real-time PCR is more accurate than pp65 antigen assay to detect HCMV infection in kidney transplant patients. We could also shown that the maximum infection of HCMV belong to spring but it was not signifi cant.
