Characterization of protein species and weighted protein co-expression network regulation of Escherichia coli in response to serum killing

Abstract
Uropathogenic Escherichia coli are responsible for approximately 90% of urinary tract infections (UTI) seen in individuals with ordinary anatomy, most of them have serum resistance. Thus, an understanding of E. coli serum antiserum is especially important to control the infectious disease caused by the bacterium. In the present study, characteristics of protein species are determined in Escherichia coli using 2-DE based proteomics. Further investigation on bacterial serum resistance indicates that more proteins with protein species are found in the bacterium in response to serum stress. Weighted protein co-expression network shows that protein species are related to topological connection as a result of protein regulation. The node protein IleS is demonstrated to contribute to serum resistance using gene-deleted mutant. The demonstration on general characteristic features of bacterial species with serum killing provides novel insights into biological significance of bacterial protein species, particularly the role in serum resistance as well as control of pathogenic E. coli.
