A) Collagen antibody induced arthritis (CAIA): On day 0, 3.2 mg anti-collagen- II antibody mix was injected i.p. in DBA/J1 mice, followed by an i.p. injection containing 25 μg LPS on day 3 with or without (no Ab) first generation ACPA or unrelated antibody (1 mg/mouse). B) Collagen induced arthritis (CIA): On day 0, 100 μg chicken type II collagen was injected intradermally at the tail base, followed by an i.p. booster with 100 μg chicken type II on day 21. On day 22 mice received an i.p. injection containing 25 μg LPS. Mice received 6 mg RhmAb2.102 or placebo (no Ab) i.p. on days 22 and 30. Mice were scored daily for signs of inflammation in their paws according to Table 1. Shown are the means ± SEM (CAIA, n=5; CIA, n=9). |
