The proposed study will investigate new treatment modalities in the field of hematopoietic stem cell transplantation. I have a vast experience in cellular therapy, in its unrefined mode of hematopoietic stem cell transplantation, as well as in more selective cell therapeutics, such as CD34+ haploidentical transplant and CD8+ modified T cells in treatment of neuroblastoma, and most recently, in the early development phases of a study investigating modification of central memory derived CD19-specific T-cell grafts in patients with childhood CD19+ ALL. In my role as the director of a pediatric BMT unit, I have the expertise, leadership and motivation necessary to successfully carry out this project. As a board certified pediatric hematologist/oncologist I have a broad exposure to pre-B cell ALL, the most common cancer diagnosis in childhood. I have served as the PI on multiple clinical trials involving all sources of hematopoietic stem cells. Currently, I am the PI on trials involving cord blood transplants, haploidentical transplants, and clinical trials utilizing novel devices employed in conditioning therapy. In my role as PI it is my responsibility to oversee the administration of the projects, including and not limited to: staffing, research protections, budget, collaboration with other investigators, and publication of peer-reviewed manuscripts from each project.  As a result of these previous experiences, I am aware of the importance of frequent communication among project members and of constructing a realistic research plan, timeline, and budget. The current application builds on my prior work, as a clinical investigator. I have a demonstrated track record of successful and productive research projects in the area of cellular therapy. I am confident that my experience has prepared me to lead the proposed project.

Tamir Miloh, MD, is the director of the liver and liver transplant program at Phoenix Children’s Hospital and Associate Professor of pediatrics at University of Arizona.  Dr. Miloh is triple board certified in pediatrics, pediatric gastroenterology, hepatology and nutrition and pediatric transplant hepatology. He earned his medical degree at Tel Aviv University and pediatric residency at St. Christopher Hospital for Children, Philadelphia, Pa. Henkie Tan, MD, PhD, is associate professor of surgery at the University of Pittsburgh School of Medicine and Children′s Hospital of Pittsburgh of UPMC. He also serves as an assistant professor of surgery of the living donor kidney transplantation program at the Thomas E. Starzl Transplantation Institute.
Dr. Tan performed the first purely laparoscopic live donor nephrectomy and developed the laparoscopic live donor nephrectomy program at UPMC where approximately 100 such operations are performed each year. He also performed the first dual kidney transplant at UPMC. Dr. Tan focuses on living donor kidney and liver transplantation and performs kidney, pancreas and liver transplants.
Dr. Tan received his combined M.D. and Ph.D. from the Medical Scientist Training Program at Loma Linda University. He received his Doctor of Philosophy in Microbiology/Immunology under the direction of Dr. Nelsen-Cannarella, a former student of Nobel Laureate Sir Peter Medawar. He completed his multi-organ transplant surgery fellowship at Johns Hopkins University School of Medicine.
Among Dr. Tan’s professional affiliations, he is a fellow of the American College of Surgeons, and a member of the American Society of Transplant Surgeons, American Society of Transplantation, the Transplantation Society, International Pediatric Transplantation Society, International Liver Transplantation Society, Central Surgical Association, Association for Academic Surgery and the Society of University Surgeons.
Dr. Tan served as the 2005-06 chair of the American Society of Transplantation Kidney and Pancreas Transplantation Committee and the 2005-07 co-chair and the 2007-08 chair of the Organ Donation Committee, among others. He has published widely in numerous transplant journals and books and is an editor of the textbook Living Donor Organ Transplantation: State of the Art. He has been nominated for America′s Top Surgeons and Best Doctors in America.

Dr. Pham is Associate Clinical Professor of Medicine and Director of Outpatient Services at the David Geffen School of Medicine at University of California Los Angeles, Kidney and Pancreas Transplant Program.  Her areas of interest include new onset diabetes after transplantation, post-transplant cardiovascular disease, and medical and urological complications after pancreas and kidney pancreas transplantation.  She is well-recognized for her work in various areas of kidney transplantation and has been invited to speak on transplant-related topics both at the national and international levels.  She currently participates in several large-scale multicenter clinical trials in the field of kidney transplantation as principal investigator and co-investigator and researches on common issues that limit kidney transplantation including the recurrence of glomerular diseases following primary renal transplantation, long-term complications following kidney transplantation, and the medico-surgical and psychosocial limitations to living kidney donation.  In addition, Dr. Pham also has an interest in hypomagnesemia and its potential pathogenic role in diabetic associated complications.

After obtaining my M.D. in China, I went to UK to pursue a Ph.D. in neuroscience. I entered the field of retinal degenerative diseases as a post-doc working with Professor Ray Lund PhD, FRS, at the Institute of Ophthalmology, UK. Our main research projects were to (i) understand the underlying pathology of retinitis pigmentosa and age-related macular disease; (ii) develop more effective and early diagnosis of these conditions; and (iii) develop therapeutic approaches to ameliorate these blinding diseases. In 2001, I moved with Professor Lund to the Moran Eye Center, UT, and was promoted as assistant professor. In 2006, I moved to Casey Eye Institute, as faculty. I established an independent research program and was award with NIH grant. Now as faculty member, I am working at Regenerative Medicine Institute at Cedars-Sinai medical center.
The outmost goal of our research is to use stem cell therapy for treating retinal degeneration.

Retinal degeneration is the leading cause of blindness in humans. There are two strategies: one is to prevent photoreceptor loss at an early stage in the disease; the other is to rebuild retinal circuitry at later stages. A promising clinical application for stem cell transplantation is in rescue therapy for degenerative diseases. We have transplanted retinal pigment epithelium (RPE) cells to replace defective host RPE; cells releasing trophic factors to increase the efficiency of dysfunctional cells; cells with multiple functions such as stem cell-derived RPE cells, mesenchyme stem cells. These cells produced rescue of photoreceptor and a range of visual functions. Histological analyses showed no evidence of tumor formation or other untoward pathology.
We also demonstrated that MSCs offer extensive preservation of photoreceptors and limit the development of secondary vascular pathology when delivered intravenously.
The outmost goal of our research is to use stem cell therapy for treating retinal degeneration.

I am currently in the second year as a tenure track Assistant Professor in the Department of Surgery at the Emory University School of Medicine. My long term is to make a relevant contribution to field of transplantation immunology and advance to the level of full Professor. It is my hope that my basic research will contribute significantly to field of B cell biology and that these advances will translate to a number of B cell directed therapies in the areas of autoimmunity, cancer and transplantation. We hope that our studies will lead to new insights into the fundamental mechanisms determining the formation of donor-specific memory B cells and plasma cells. We anticipate that these studies will generate novel gene or pathway candidates for potential study and therapeutic targeting.

Success in kidney transplantation has resulted from control of T-cell-mediated acute rejection. The use of increasingly potent T-cell directed immunosuppressants has led to a reduction in graft loss from acute rejection. However, little has been done to improve the fate of patients who possess pretransplant donor-specific antibody (DSA), and no proven therapies exist to specifically prevent DSA formation post-transplant. As such, sensitized patients are the most challenging patients to transplant. While methods to detect and characterize DSAs are clearly useful, the B-cell subsets that produce DSAs, or more importantly sustain their production are poorly characterized with regard to phenotype, survival mechanisms, and anatomical locations. DSA-mediated disease is a growing problem in clinical transplantation. We are interested in understanding the cellular and molecular mechanisms that regulate DSA production.To this end, we hope that enhanced knowledge of the basic biology of B-cell activation and participation in multi-cellular responses to alloantigens will lead to novel therapeutic avenues.

After my Medical School studies at Leeds I began a career in Academic Surgery in Oxford, specialising in transplantation and vascular surgery. My most significant contribution was being the first to show that it was possible to isolate human islets of Langerhans from the human pancreas in sufficient numbers to contemplate transplantation. That work has finally come to fruition in 2008 with the setting up of an NHS funded, nationwide islet transplantation program to treat patients with particulalry severe diabetes by islet transplantation, Oxford being one of two UK islet isolating centres. The challenge is now to set up a successful continuing clinical islet transplant programme.

The barriers of immune rejection and auto-immune disease are crucially important to the long-term success of cellular transplantation, and arising from my reading of past and present research in these fields I have proposed a fundamental alteration in the interpretation of the role of MHC (Major Histocompatibility Complex) molecules, which I have named MHC Based Suppression (MBS). {Link to Oxford Research Archive (http://ora.ouls.ox.ac.uk:8081/10030/989)}.

A further interest has been the development of a computerised timetabling system for Medical School teaching, which also gives the opportunity to feed back the students opinion of the previous teaching. By adding in the concept of notional credits for teaching performed it is possible to identify and reward those teachers who deliver excellence. This system is running in one medical school (not UK) and is in the process of being introduced to others. I declare a financial interest in this development.

In Oriel, my main contribution is to provide Gross Anatomy teaching to first-year Medical Students. To improve the teaching efficiency I have introduced computer driven projection of "dissectable anatomy" and also synchronous dual projection to show radiology (provided by a Consultant Radiologist) of the points of interest.

Dr. Jardine graduated in Physiology & Medicine from the University of Glasgow in 1984. He undertook postgraduate research training as a Clinical Training Fellow in the MRC Blood Pressure Unit, where he pursued studies on the physiology, pharmacology and therapeutic potential of natriuretic peptides. He trained initially in Endocrinology and switched to Nephrology in 1990. After clinical training in Nephrology in Glasgow, he was a Lecturer/ Senior Registrar in Aberdeen & Inverness, and worked on proximal tubular cells and the actions of nitric oxide. After graduation he entered academic practice including basic science and clinical training within liver transplantation center of Zhejiang University and Queen Mary Hospital of Hong Kong University. From August 2010 he was appointed the head of Dept. Hepatobiliary Pancreatic Surg,First Affiliated Hospital of Hainan Medical University.
He strongly experienced in clinical and fundamental work of hepatobiliary surgery and liver transplantation. The Institute is associated with the Lateran University in Rome and is accredited in Australia to conduct graduate programs (Grad Dip, Masters and PhD) in Theology and Bioethics. He is a Bioethicist and currently chair of the National Health and Medical Research Council Working Committee on Commercialization of Human Tissue. He has strong interest in transplantation. He was also a member of thre NHMRC Committee on ART which produced guidelines for stem cell resesarch. He recently published an article on the death issue for the Journal of Bioethics. Nick has served on numerous government committees including the Australian Health Ethics Committee where he was a deputy chair and chaired committees on the unresponsive state and on the commercial use of human tissue. In 1982 he was Australias first hospital ethicist at St Vincents Hospital where he was Director of Bioethics. He has taught medical students, nurses and others for thirty years and after leaving St Vincent’s has been a consultant serving the Office of the Australian Prime Minister, the Australian Minister for Health, the US Congress, the Victorian Minister for Health, the German Federal Department of Health and Welfare, UNESCO and the Australian Catholic Bishops. In 2009 he was appointed a papal knight by Pope Benedict XVI for his work in Bioethics and for the Catholic Church.

Nick is chronically ill with renal disease and has been on dialysis for the past nineteen years. The latter led to coronary disease and he had some bad luck with a failed coronary bypass operation in 2004 after which he has had fifteen angioplasty procedures and the placement of eight stents. He is married to a GP, Dr Mary Walsh, and they have four children. The underlying disease has also caused vision disability and neuropathy which means that he now uses dictation software for much of his output. Despite that he has maintained his research active status and currently has five articles forthcoming in peer reviewed journals and is completing the co-authorship of a guide to current ethical issues.

He had his medical education at the University of Ibadan, Nigeria, graduating with MB, BS in 1987. He completed US residency in Internal Medicine at Michael Reese Hospital/University of Illinois at Chicago before going to the University of Vermont/Fletcher Allen Health Care as a Nephrology Fellow. Following his training in Nephrology here, he went to the Oregon Health & Science University as a fellow and Clinical Instructor of Medicine in Kidney Transplantation. He returned to Cyprus as the only certified Transplant Nephrologist. He worked as a Consultant Transplant Nephrologist at the Paraskevaidion Transplant Center and then after agreement with the Ministry of Health, he joined the New Organ Transplantation Unit of the Nicosia General Hospital. He is active member of the American Society of Transplantation, the Americam Society of Nephrology and the European Society of Organ Transplantation. His research interest includes acute rejection markers and Pancreas Transplantation.
